The former Robin and Batgirl will appear as Nightwing in the DC Comics series.
She will join Batman in his debut in the title in Smallville #5.
Nightwing has traditionally been the role filled by Dick Grayson following his graduation from his position as the original Robin.
"Bruce can be somewhat of an angry man," writer Bryan Q Miller told TV Guide.
"Stephanie's personality is so can-do and unsinkable and bright, so it's very much on purpose on Bruce's part that he has a good cop going out on patrol with him every night."
Miller previous wrote Stephanie as the star of the pre-New 52 Batgirl series. He promised that the comic would explain why she has taken up the mantle of Nightwing.
The Smallville comic continues the story of the recently-ended television show.
Smallville Season 11 #5 will be released in September.
