The announcement was accompanied by the same trailer introduced by creators Matt Stone and Trey Parker during Microsoft's E3 press conference.
It was also revealed that players who pre-order the game will receive the 'Good Times With Weapons Pack', a collection of bonus weapons based on the popular South Park episode.
Xbox 360 users, meanwhile, will also receive the 'Mysterion Superhero Pack', which features new items and costumes, including the 'Dagger of Cthulhu' from the 'Mysterion Rises' episode.
The Xbox 360 also benefits from Kinect support, while the first three add-ons will be timed Xbox exclusives.
Kinect features include voice commands for taunting enemies, launching attacks and insulting Cartman.
Originally scheduled for late 2012, South Park: The Game sees a new kid arrive in South Park who takes part in a live-action role playing game with the other kids that eventually becomes an adventure of its own.
It is developed by Fallout: New Vegas studio Obsidian Entertainment and written by show creators Trey Parker and Matt Stone.
Earlier this year, Obsidian Entertainment was struck by layoffs that included members of the South Park team and caused the studio to cancel an unannounced title.
South Park: The Game will be available for Xbox 360, PlayStation 3 and PC.
