The upcoming drama - based on the BBC series - will focus on the lives and relationships of four female friends.
The Artist star Miller has won a recurring role on the show, Entertainment Weekly reports.
Her character - the mother of Sam Gray (Erik Stocklin) - is unaware that her late husband was having an affair with Karen Rhodes, played by Lost actress Yunjin Kim.
The 48-year-old Miller is known for roles in such films as 1990's Kindergarten Cop and 1993's Carlito's Way, while her television credits include TNT's Men of a Certain Age.
Mistresses will premiere on ABC in May 2013.
