The 40-year-old played CIA agent Sydney Bristow on the ABC drama - created by JJ Abrams - between 2001 and 2006.
"I think JJ Abrams has got his hands full," Garner told Fretts on Film. "I don't see him turning around and rebooting Alias any time soon."
However, the actress added: "If he was involved, I'm sure the rest of us would sign right up. We had a blast making that show and we're all still super close, so I'm sure you would find an eager group of participants right there."
Garner's most recent credit is Disney's The Odd Life of Timothy Green while Abrams is an executive producer on Fringe, Person of Interest and new NBC drama Revolution.
In 2010, it was reported that ABC was interested in producing an Alias remake, but series star Michael Vartan later dismissed the reports as "total rumour".
"If they do redo it and they don't bring back the original cast, that would be weird," he argued.
